Asymmetric copper-catalyzed conjugate additions of organometallic reagents in the syntheses of natural compounds and pharmaceuticals.
Access to enantiopure complex molecular structures is crucial for the development of new drugs as well as agents used in crop-protection. In this regard, numerous asymmetric methods have been established. Copper-catalyzed 1,4-additions of organometallic reagents are robust C-C bond formation strategies applicable in a wide range of circumstances.
This review analyses the syntheses of natural products and pharmaceutical agents, which rely on the application of asymmetric Cu-catalyzed conjugate additions of various organometallic reagents. A wide range of available organometallics, e.g. dialkylzinc, trialkylaluminum, Grignard, and organozirconium, can now be used in conjugate additions to address various synthetic challenges present in targeted natural compounds. Furthermore, efficient catalysts allow high levels of stereofidelity over a diverse array of starting Michael acceptors.

Evaluation of a new thromboplastin reagent STA-NeoPTimal on a STA R Max analyzer for the measurement of prothrombin time, international normalized ratio and extrinsic factor levels
Introduction: We aimed at evaluating the performance of a new prothrombin time (PT) reagent (STA-NeoPTimal) with two other PT reagents (STA-Neoplastine R and STA-Neoplastine CI Plus) and the reference PT reagent used in our laboratory (ReadiPlasTin).
Methods: Evaluation consisted in intra- and interassay precision assessment, determination of sensitivity to unfractionated heparin (UFH) or enoxaparin in spiked samples and to direct oral anticoagulants (DOACs) in patients (n = 43). Method comparison of the 4 PT reagents, factor II, V, VII and X assays was tested on normal (n = 20) and abnormal samples: VKA (n = 47), preoperative (n = 23), liver failure (n = 12) and burned patients (n = 37).
Results: Analytical performance met manufacturers’ criteria for all reagents. All PT reagents gave correlation coefficients >0.8 and even >0.9 in many situations.
In some VKA samples, differences ≥ 0.5 INR units were found in samples within and above therapeutic ranges. For burned patients, PT correlations were good but with some minimal bias (<5.0%) while factor assays gave very consistent results (R > .8 and mainly >0.9). As expected, poor responsiveness of the PT to DOAC concentrations was observed with all four assays.
Conclusion: The STA-NeoPTimal showed comparable performance to ReadiPlasTin, making it suitable for VKA control, detection of factors II, V, VII, X deficiency and assessment of liver disease coagulopathy.
However, for patients receiving VKA, some significant differences were observed. We confirmed the inability of the PT assay to detect residual DOAC concentrations. Finally, burned patients results showed that recombinant thromboplastins were less sensitive to factor deficiencies in comparison to extraction thromboplastins.
An Experimental and Theoretical Study on the “2,2′ Bipiridyl Ni Catalyzed” Hirao Reaction of >P(O)H Reagents and Halobenzenes; A Ni(0)→Ni(II) or a Ni(II)→Ni(IV) mechanism?
The P-C couplings between >P(O)H reagents and halogenoarenes may take place either by Pd- or Ni-catalysis. However, the latter version has been less studied. It was found by us that the reaction of Ph2P(O)H or PhEtOP(O)H with PhX (X = I and Br) in the presence of NiCl2/Zn as the precursors for the assumed Ni(0) complexant together with 2,2′-bipyridine as the ligand took place only with PhI at 50/70 °C. M06-2X/6-31G(d,p)//PCM(MeCN) calculations for the P-C coupling of Ph2P(O)H and PhX revealed a favorable energetics only for the loss of iodide following the oxidative addition of PhI on the Ni(0) atom.
- However, the assumed transition states with Ni(II) formed after P-ligand uptake and deprotonation could not undergo reductive elimination meaning a “dead-end street” in the mechanism expected. Hence, it was assumed that the initial complexation of the remaining Ni2+ ions with 2,2′-bipyridine may move the P-C coupling forward via a Ni(II)→Ni(IV) transition.
- This route was also evaluated by calculations, and the mechanism explored was justified by preparative experiments carried out using NiCl2 and bipyridine, but in the absence of Zn. There was no need for Zn, when bipyridine was applied as the N-ligand, moreover PhBr could also be a suitable starting material at higher temperatures.
- Hence, the generally accepted Ni(0)→Ni(II) route was refuted by us, confirming, surprisingly, the generality of the Ni(II)→N(IV) protocol, either in the presence of bipyridine, or using the excess of the >P(O)H ⇌ >P-OH reagent as the P-ligand.
- The results of the calculations on the complex forming ability of Ni(0) and Ni(II) with 2,2′-bipyridine or the tautomeric >P-OH form of the P-reagents were in accord with our mechanistic proposition. Our study also means the clarification of the contradicting and sometimes uncertain literature data published in the last decade.
